Unit 1

Unit 1: Microbiology Computer Use and Lab Safety By Karen Bentz, Patricia Wilber, Heather Fitzgerald, and Andrea Peterson, 2022Creative Commons Attribution-NonCommercial 4.0 International License.Introduction to Tablet Computers and the online Lab Manual. In order to prevent microbes from going home on your belongings, you will be assigned a tablet computer to be used in lab for the term for all notes taken in this class, or at your instructor’s discretion, you can bring your own phone or tablet (not a laptop). However, these units must be kept in a plastic bag at all times and the bag will remain in the classroom. You will NOT bring anything else with you to class nor will you take materials home. You are responsible for taking care of your tablet, returning it to its proper place in the storage cabinet when you are done using it for the day, and for reporting any issues with the tablet to your instructor. LAB MANUAL: This is the link for the free online lab manual: is a desk top icon for the lab manual on the class room tablets.VIDEO LINKS: Some of the Units have videos. The video links are embedded in the text of the Units and are in stand-alone form on the book website. You should view the videos at home BEFORE lab. There will not be enough time to view them as you try to complete the activities each day in lab.USING A TABLET:Obtain a tablet from the storage cabinet.Record your name and the number of your tablet on the sign out sheet provided by your instructor. YOU WILL USE THE SAME TABLET EVERY CLASS! This is important because the tablet will “remember” you.Carefully follow the directions on the handout for using your HP-ElitePad 1000 tablet. Maximize your Unit 1 file and continue with the lab safety activities.Introduction to Microbiology Lab SafetyYour safety is the number one priority in the microbiology lab!39719251270000In this class you will be working with living microorganisms that have the capacity to cause illness to you and your family. It is of utmost importance that you familiarize yourself with these potential pathogens and the lab regulations you must follow when dealing with these organisms. Please read the information below, and then sign the safety contract to indicate that you understand thisinformation and will abide by these safety rules. When completed please give the signed contract to your instructor. Most of the bacteria that you will work with in this lab are classified by the Centers for Disease Control (CDC) and the American Society for Microbiology (ASM) as Biological Safety Level 1 (BSL) and BSL 2 organisms (1). This means most of the bacteria you will be using can pose a small risk of infection to you, and are at low risk for causing illness in the general community. When good microbiological techniques are used, these organisms rarely cause serious disease, and effective treatment for laboratory- acquired infections is available (2). However, if you do not adhere to these methods it is always possible to contract an illness, as this lab deals with living microorganisms. If you have special medical concerns, please bring these to your instructor’s attention when you fill out the medical safety contract.BSL-1 Low level of pathogencity. BSL-1 organisms you will work with in this lab include: Citrobacter freundii, Escherichia coli, Serratia marcescens, Priestia megaterium, Bacillus subtilis, and Staphylococcus saprophyticus.BSL-2 May cause illness; higher level of pathogenicity than BSL-1. BSL-2 organisms you will work with in this lab include: Aggregatibacter aphrophilus, Klebsiella pneumoniae, Proteus vulgaris, Pseudomonas aeruginosa, Shigella flexneri, Staphylococcus aureus, Enterococcus faecalis, Streptococcus pneumoniae, Streptococcus pyogenes and Streptococcus oralis.Safety Guidelines for Microbiology LabTo minimize the risk of exposure and infection, you are required to follow these guidelines:Use common sense in the lab. If you see a potential hazard, let your instructor know.No one other than registered students are allowed in lab. This includes children and pets. With proper handling and attention to procedure and protocol, risk of illness due to exposure to bacteria in Microbiology Lab is almost non-existent. ?However, students who are pregnant or become pregnant during the term, as well as students with compromised immune systems, should seek advice from their medical provider(s) and inform their instructor(s) if any concerns are identified.Student personal items will be limited. Once live cultures are in use in the lab students are not allowed to have any of their personal items (pens, wallets, etc.) with them in lab. Personal electronic devices must be in plastic bags at all times and should be wiped with a clean Kimwipe and lens cleaner at the end of the lab prior to leaving the room. They can rest in a drawer after being cleaned but should not be set on the countertops without a baggie.Food (to include gum and lozenges) and drinks are never allowed in lab. Students will need to store their belongings in the designated area of the lab or in lockers provided. There will be a specific procedure for safely taking lab notes during class in the event that the computers do not work.Classroom materials (books, pens, dirty lab notes will remain in the class to minimize transfer of microorganisms to the community.Students will be required to wear cloth lab coats in lab. Disposable or hip-length coats are not allowed. Lab coats must be of a length to reach to the wrist and covered to the knee when sitting (generally this is a ? length coat). We can provide used (but clean!) coats to most students. Students may also bring in their own coats, but the coats must remain in lab until the end of the term when they will be sterilized and then returned to the student. Underneath the coats students should wear pants and/or skirts that go at least to mid-calf. Shorts, capris and any other legwear that does not go at least past the knee is not allowed. Instructors may require more stringent clothing guidelines and students are expected to follow any additional expectations their instructor chooses for their sections.Students must wear closed- toed and closed heel shoes. No flip flops. No clogs. It is best to have a separate set of lab shoes and home shoes to prevent carrying any bacteria that may contaminate the floor from being tracked back into your home. Students with long hair must tie their hair back. Long dangling jewelry, scarves or anything that is loose and may touch the lab bench should not be worn. Hats should not be worn during lab.Students will wear nitrile gloves while dealing with any surfaces that may be contaminated with bacteria. Do NOT keep them on when opening the door to the classroom or when handling materials that may return to your house (your eyeglasses etc.). Discard used gloves in the biohazard bucket. To minimize student exposure to aerosols that may be generated, students will wear face shields when dealing with live organisms (inoculating, adding reagents, etc.). Students will not be allowed to participate in lab if they are not dressed in appropriate lab clothing.Students should wash their hands with soap and water and dry them with paper towels before and after the lab.The lab benches will be outfitted with high grade disinfectant and students will clean the benches before and after use with these chemicals. All waste will be disposed of in the appropriate containers. Students should be aware of the location of the following lab safety features:Lab exitsFirst aid kitFire extinguisher(s)Shower and/or eye washLAB ACTIVITY USING YOUR CAMERAFind all of these items: lab exits, first aid kit, fire extinguishers, shower and eyewash. Using your tablet or personal device, photograph (main screen, camera icon), two of these items. Insert the pictures below and label them. To insert them, choose “INSERT” from the Tabs at the top of the page. Choose “Pictures”. Click on the image you want to insert. Click “Insert”.Label your pictures by choosing “INSERT” on the top of the page, then “Text Box” on the top right. BE SURE TO INSERT RATHER THAN COPYING AND PASTING PHOTOS. INSERTING SAVES A LOT OF SPACE.After posting and labelling your pictures, proceed to the next Section: “Microbiological Waste Disposal”Insert your two pictures in the space below, and then label them. If you left click the picture, you can change the format of the picture to make them easier to manage. Microbiology Waste DisposalThe materials you use in the microbiology lab need to be disposed of properly. The following is a summary of where various items can be safely discarded.
