NIH/NIA MRI, Genetic and Cognitive Precursors of AD and Dementia Role: Investigator
The goals of the study are to relate risk factors for Alzheimer’s Disease (AD) and dementia in the Framingham Offspring and Omni cohorts to changes in brain morphology measured by Magnetic Resonance Imaging (MR) and cognitive performance. Longitudinal data on change in MRI and cognitive measures will enable us to further explore the hypothesis that CVRF prospectively ascertained during mid-life play an important role in progressive brain injury leading in turn to progressive decline in cognition, increasing the likelihood of mild cognitive impairment (MCI) or dementia in later life. Finally, we will combine extensive genetic information in younger individuals with a documented family history of dementia to explore potential early changes in brain structure and cognition that may be under genetic influence.

Epidemiology of Dementia in the Framingham Study Role: Investigator
The goals of the study are to expand cognitive surveillance efforts to the older FHS Offspring and multi-ethnic OMNI cohorts permitting us to explore the familial occurrence of AD and dementia and to relate mid-life habits and risk factors to incidence of AD and other types of dementia. More than 420 of these Offspring and OMNI subjects have volunteered as brain donors, and we plan to relate vascular pathology in the brain at autopsy to mid-life vascular risk factors measured over several decades. This study will permit us to address a key question of whether vascular risk factors, especially those present at midlife, increase the risk of clinical and pathological dementia generally, and AD specifically.

Precursors of Stroke Incidence and Prognosis Role: Investigator
The study will further the understanding of the epidemiology of stroke through identification of newer risk factors and genetic influences. Clinical and subclinical strokes will be studied utilizing the rich multi-generational family-based structure. Clinical and subclinical strokes will be related to documented clinical stroke in parents and siblings to assess the impact of stroke risk factors depending on family history of stroke.
