This research involved determining the life-cycle of a novel protozoan

parasite that uses viperid snakes in the genus Vipera as definitive hosts and

Current Research:

Protein biochemistry of Cryptosporidium

Our laboratory is examining the cytochemistry, biochemistry and molecular biology of

Cryptosporidium parvum in vitro and in vivo, and attempting to characterize parasite

ligand/host cell receptor interactions involved in host cell recognition and penetration.

This research should help characterize the molecular mechanism by which the parasite

gains access to a susceptible host cell. We have also examined UV-repair

mechanisms in this parasite, and have cloned most of the genes in this pathway.

dsRNAs of Cryptosporidium

Previously, we described the presence of 2 unique, linear, dsRNAs located within

the cytoplasm of sporozoites of C. parvum. These dsRNAs are packaged as viruses

and are transmitted vertically within the parasite. All indications suggest these to be

unique only to several closely related members of the genus. We are currently

examining methods by which the dsRNAs replicate, undergo translation, and can be

employed in species specific detection of the parasite in environmental samples.

In vitro cultivation of coccidia

Our laboratory is exploring mechanisms whereby development of Cryptosporidium can

be enhanced in vitro. These studies include the trial and error of select medium

supplements, as well as using alternative host cell types. Recent work involving

cultivation in ovo have revealed C. parvum to develop well in chicken embryos.

award represented a graduate scholarship which would have covered my first

semester in graduate school at Toronto University. I declined the award because

I decided to attend Auburn.

"In ovo techniques for cultivation of coccidia." A two day session presented by Dr. P. L. Long,

Department of Poultry Science, University of Georgia, Athens (July, 1982) which focused

on techniques on growing protozoa in chicken embryos.

"Identification of oocysts and intestinal lesions of chicken and turkey coccidia." A two session

presented by Dr. S.A. Edgar and Hoffman-LaRoche pharmaceuticals, Department of Poultry

Science, Auburn University (April, 1984) which focused on identifying protozoan parasites

within the intestine of poultry based on gross lesions.

"Cells in Space-II; the effects of microgravity on biological culture systems and potential for

commercialization in space." Workshop and Conference, presented by NASA/Ames

Research Center at Saint Francis Retreat, San Juan Bautista, California (31 Oct.-4 Nov. 4,

1988) which focused on new technology available for growing cells in space.

"Cryptosporidium workshop." Sponsored by the U.S.E.P.A. and the Center for Environmental

Epidemiology, Graduate School of Public Health, University of Pittsburgh (10-11 Jan.,

1989). This meeting focused on new developments in our knowledge of cryptosporidiosis.

"Discovery and development of therapeutic agents for opportunistic infections associated with

AIDS." Division of AIDS and Division of Microbiology & Infectious Diseases, NIH,

Bethesda (11-12 Sept., 1989). This meeting helped summarize the extent of what was

known about cryptosporidiosis, and revealed previously unpublished clinical trial data.

"Prospects for in vitro growth and propagation of Cryptosporidium parvum." Division of AIDS

and related diseases, therapeutics branch, NIH, Bethesda (28 Sept., 1995). This workshop

focused on new techniques with potential for growing C. parvum more efficiently in vitro.

(NIH workshop on Cryptosporidium chemotherapy: clinical studies, animal models, and new

leads,( Sept. 18-19, 1997, Bethesda, MD. This workshop presented the data on the

newest pharmaceuticals that were being developed against the parasite.

(Cryptosporidium disinfection workshop - towards a standardized experimental design for viability

and inactivation studies.( 11-12 July 1999, Tadley Court, Basingstoke, Hampshire UK.

This workshop involved developing a series of guidelines and standards which will be

required of all investigators receiving grants in the United Kingdom when they study

disinfection and control strategies of Cryptosporidium.

(Cell culture workshop, Water Quality and Technology Conference.( 31 October-02 November,

1999, Tampa Convention Center, Tampa, FL. This workshop was designed to train

technicians in the water industry on various in vitro methods used to identify and assay

for Cryptosporidium in public water.

(Wildlife capture, immobilization, handling, and release techniques.( 13-14 November 1999,

Kansas State University. The workshop, taught by Dr. Mark Johnson of the University of

Montana, provided basics on different wildlife handling techniques, chemical

immobilization, and capture and release.

Graduate Students and Post-graduate Fellows:

Keith M. Woods (Post-doctoral fellow. 1994-present). Keith was instrumental in developing the

first colorimetric ELISA for measuring the extent of development of the parasite in vitro.

He also develops and maintains hybridomas in the laboratory.

Peter Chung (PhD, 1997-2004). Processing of beta-glucan synthetase in Cryptosporidium

parvum. Peter determined the method by which the parasite translates and processes

beta-glucan sythetase, which is originally transcribed as a large, multi-component precursor

protein prior to being broken down into multiple functional enzymes.

Nikolai Khramtsov (Post-doctoral fellow. 1992-2000). Nikolai developed over 20 genomic

libraries of Cryptosporidium parvum or various other members of the genus, and most

recently worked on the molecular biology of a unique dsRNA virus found within C. parvum.

He has taken a job with CuraGen Inc. in Connecticut.

Michael V. Nesterenko (Post-doctoral fellow. 1993-2000). Michael developed novel methods

of purifying surface proteins of Cryptosporidium parvum. He has returned to Russia and is

employed in private industry.

Gabriela Dontu (PhD, 1994-2000). Stage specific expression of Cryptosporidium parvum

genes. Her research involved determining which parasite genes are being switched on

during the infection process. Gabriela is working as a post-doctoral fellow at the University

of Michigan cancer research center, Ann Arbor.

Jamie Kasper (MS, 1999-2000). The histone deacetylase gene of Cryptosporidium parvum.

Jamie collaborated with researchers at Merck Inc. studying the susceptibility of

recombinant parasite histone deacetylases to a new class of anti-parasitic agents. She took

a position with a biotechnology company in Dallas.

Leasa Brown (MS, 1996-1998). The histone deacetylase gene of Cryptosporidium parvum.

Leasa initiated work on the histone deacetylases of C. parvum. She received a job offer

from Midwest Research Institute prior to completing her thesis.

Angela Lambley (M.S., 1995-present). Protein kinase of Cryptosporidium parvum. Angela

also received a job offer prior to degree completion. Her research focused on sequencing

and understanding motiffs in a key protein kinase in Cryptosporidium. She is currently

writing her thesis while working as a technician in the Division of Biology at KSU.

Michael Tilley (Post-doctoral fellow. 1991-1993; PhD 1986-1991). Immunoparasitology; in vitro

cultivation. An analysis of Cryptosporidium parvum sporozoites using lectins,

125I surface labeling, and monoclonal antibodies. Mike also developed methods to cultivate

coccidia in vitro and developed monoclonal antibodies as probes for proteins of

Cryptosporidium parvum. He received a Hitchings-Elion post-doctoral fellowship from

Burrough(s Wellcome to study for 2 years in England, and is currently working as a scientist

at the USDA grain marketing center in Manhattan KS.

Brenda S. Oppert (Post-doctoral fellow. 1993). Protein biochemistry and molecular biology.

Brenda characterized the superoxide dismutase in Cryptosporidium. She is currently

working as a scientist at the USDA grain marketing research center in Manhattan, KS

Dennis Blunt (PhD candidate, 1993-1995). Pulse field electrophoresis of of Cryptosporidium

parvum. Dennis spent time in the laboratory for several years and studied the genome

composition of Cryptosporidium by pulse field electrophoresis. He eventually left the PhD

program to work in private industry.

Ralene Mitschler (PhD, 1989-1994). Lipid biology of Cryptosporidium parvum and Eimeria

nieschulzi (Apicomplexa). Ralene characterized the amounts and types of lipids in two

different parasite species. She received a post-doctoral position at Stanford University, and

is currently an Assistant Professor at Western Maryland University.

Dianne B. Brillhart (MS, 1990-1993). Sero-prevalence of Lyme disease antibodies in Peromyscus

leucopus, Peromyscus maniculatus and Odocoileus virginianus, and distribution of potential

tick vectors, in Kansas. Diane was the first student in decades to initiate a state wide survey

in Kansas of the species of ticks present. She also examined the seroprevalence of Lyme

disease in wildlife in the state. She went on to obtain her PhD at the University of

Kentucky and is currently in a post-doctoral position at Kansas State University.

Mark Eggleston (MS, 1990-1993). Development of a 3H-uracil incorporation assay to monitor in

vitro development Cryptosporidium parvum. Mark developed a tritiated uracil incorporation

assay to quantitate development of Cryptosporidium in cell cultures. Mark went on to

Medical school at Creighton University and is now a physician.
