Izbornom ve?u Biolo?kog fakulteta Univerziteta u BeograduPRIVATE Na I redovnoj sednici Izbornog ve?a Biolo?kog fakulteta, Univerziteta u Beogradu, odr?anoj 14. oktobra 2016. godine, odre?eni smo u Komisiju za pisanje Izve?taja o prijavljenim kandidatima za izbor jednog vanrednog profesora za u?u nau?nu oblast Biologija mikroorganizama na Katedri za mikrobiologiju u Institutu za botaniku i Botani?koj ba?ti “Jevremovac” Biolo?kog fakulteta Univerziteta u Beogradu, na odre?eno vreme od 5 godina.Na konkurs objavljen 26. oktobra 2016. godine u listu “Poslovi” prijavio se jedan kandidat, dr Biljana Nikoli?, docent na Katedri za mikrobiologiju Biolo?kog fakulteta Univerziteta u Beogradu. Na osnovu prilo?ene dokumentacije, kao i na osnovu li?nog uvida u rad kandidata, podnosimo Izbornom ve?u slede?iI Z V E ? T A J1. BIOGRAFSKI PODACIdr Biljana Nikoli? ro?ena je 14. novembra 1971 godine u Ni?u. Osnovnu i srednju ?kolu je zavr?ila u Ni?u. Na Biolo?ki fakultet, Univerziteta u Beogradu, upisala se ?kolske 1992/93. Diplomirala je 1998. godine sa prose?nom ocenom 9,75. Diplomski rad iz oblasti mikrobiologije, pod naslovom ″Antimutageni efekat ekstrakta ?alfije (Salvia officinalis L.) na Salmonella typhimurium″ odbranila je sa ocenom 10. Poslediplomske studije na Biolo?kom fakultetu, smer Biologija mikroorganizama, koje je upisala ?kolske 1999/2000, zavr?ila je sa prose?nom ocenom 10. Magistarski rad pod naslovom ″Konstrukcija bakterijskih testova za detekciju antimutagenog potencijala antioksidanata″ uradila je na Katedri za mikrobiologiju Instituta za botaniku i Botani?ke ba?te “Jevremovac” Biolo?kog fakulteta Univerziteta u Beogradu, pod rukovodstvom prof. dr Jelene Kne?evi?-Vuk?evi? i odbranila ga 2005. godine. Doktorsku disertaciju pod naslovom "Efekat kamfora, eukaliptola i tujona na reparaciju DNK i mutagenezu u prokariotskim i eukariotskim ?elijama", koju je uradila pod rukovodstvom prof. dr Branke Vukovi?-Ga?i?, odbranila je 1. novembra 2011. godine. Na Katedri za mikrobiologiju Instituta za botaniku i Botani?ke ba?te “Jevremovac” Biolo?kog fakulteta Univerziteta u Beogradu, radi od 2000. godine i to najpre kao stipendista Republi?kog zavoda za tr?i?te rada na radnom mestu asistenta-pripravnika (2000-2003), a zatim kao istra?iva? pripravnik (2003-2005). Za asistenta na Katedri za mikrobiologiju Instituta za botaniku i Botani?ke ba?te “Jevremovac” Biolo?kog fakulteta Univerziteta u Beogradu izabrana je 2005. godine, a reizabrana 2009. U zvanje docenta za u?u nau?nu oblast Biologija mikroorganizama na istoj katedri izabrana je 2012. godine.dr Biljana Nikoli? je ?lan Dru?tva geneti?ara Srbije, Udru?enja mikrobiologa Srbije i Srpskog biolo?kog dru?tva, kao i Evropskog dru?tva za mutagenezu agensima iz okoline (European Environmental Mutagen Society - EEMS) i Federacije evropskih mikrobiolo?kih dru?tava (Federation of European Microbiological Societies - FEMS). 2. NASTAVNO - PEDAGO?KI RADU periodu 2000-2011. godine, dr Biljana Nikoli? u?estvovala je u izvo?enju nastave na osnovnim i poslediplomskim studijama na studijskim grupama Biologija, Molekularna biologija i fiziologija i Ekologija i za?tita ?ivotne sredine, na Biolo?kom fakultetu Univerziteta u Beogradu, najpre u zvanju asistenta-pripravnika, a zatim i u zvanju asistenta. Tako?e ?е u?estvоvala u izvo?enju prekti?ne nastave na studijskoj grupi Biohemija na Hemijskom fakultetu Univerziteta u Beogradu. U reformisanim studijskim programima akreditovanim na Biolo?kom fakultetu Univerziteta u Beogradu, dr Biljana Nikoli? u?estvuje u pripremi programa i izvo?enju prakti?ne nastave na osnovnim, master, specijalisti?kim i doktorskim studijama.Nakon izbora u zvanje docenta, dr Biljana Nikoli? je u?estvovala u realizaciji nastave na vi?e predmeta osnovnih akademskih studija, master studija, specijalisti?kih i doktorskih studija, a koje organizuje Katedra za mikrobiologiju. Za predmete Mikrobiologija i Mikrobiolo?ki praktikum na osnovnim akademskim studijama, predmete Metode u mikrobiologiji, Ekogenotoksikologija i Genetika bakterija i bakteriofaga na master studijama, predmete Metode u mikrobiologiji (specijalisti?ki kurs), Mikrobiologija - vi?i kurs i Primenjena mikrobiologija na specijalisti?kim studijama, kao i predmet Metode u mikrobiologiji - vi?i kurs na doktorskim studijama, kandidat je pripremila dopune nastavnih programa. Tako?e je sa preuzetim programom u?estvovala u realizaciji nastave na predmetima Osnove genotoksikologije (osnovne studije) i Antimutageneza i hemoprevencija (doktorske studije). Koautor je pomo?nog univerzitetskog ud?benika “Mikrobiolo?ki praktikum“, koji je 2014. godine objavljen u izdanju Biolo?kog fakulteta Univerziteta u Beogradu. Praktikum se koristi u realizaciji prakti?ne nastave na nekoliko predmeta (Mikrobiolo?ki praktikum, Metode u mikrobiologiji, Metode u mikrobiologiji – vi?i kurs). Prema studentskim anketama, kvalitet nastave koju je dr Biljana Nikoli? dr?ala u svojstvu docenta je vrednovan ocenama koje su se kretale u intervalu 4,30-4,91, a prose?na ocena u prethodnom izbornom periodu iznosi 4,71. U cilju unapre?enja sopstvenih pedago?ko-didakti?kih ve?tina, poha?ala je seminare za obuku nastavnog osoblja Univerziteta u Beogradu – TRAIN, na teme “Ve?tine dr?anja efektivnih prezentacija” i “Umre?avanje i timski rad”. Pored realizacije nastave na Biolo?kom fakultetu, dr Biljana Nikoli? je u?estvovala u realizaciji edukativno-istra?iva?kog programa u istra?iva?koj stanici Petnica za talentovane u?enike srednjih ?kola, као i u takmi?enjima Nauka mladima, kao ocenjiva?. Kao predava? u?estvovala je u akreditovanom programu Biologija mikroorganizama za stru?no usavr?avanje nastavnika i profesora biologije, zaposlenih u osnovnim i srednjim ?kolama. Tokom 2013. godine dr Biljana Nikoli? je dr?ala predavanje po pozivu u okviru Centra za predava?ku delatnost Kolar?eve zadu?bine, na temu ?Virusi, model globalnog parazitizma“. Dr Biljana Nikoli? je kao komentor rukovodila izradom jedne doktorske disertacije. Nakon izbora u zvanje docenta rukovodila je izradom 11 master radova (od toga 5 samostalno), kao i 4 diplomska rada, a tako?e u?estvovala u komisijama za odbranu 2 dokorske disertacije, 4 master rada i 1 specijalisti?kog rada. Tokom celokupne profesionalne karijere bila je i ?lan komisija za odbranu 13 diplomskih radova. NAU?NO-ISTRA?IVA?KI RADdr Biljana Nikoli? se od 2000. godine bavi nau?no-istra?iva?kim radom u oblasti mikrobiologije i genotoksikologije na Katedi za mikrobiologiju Biolo?kog fakulteta Univerziteta u Beogradu. Posebno interesovanje je pokazala za izu?avanje mehanizama reparacije DNK, SOS odgovora, mutageneze i antimutageneze kod prokariota. Konstrukcija bakterijskih testova za detekciju antimutagenog potencijala antioksidanata bila je predmet njene magistarske teze, a detekcija bioantimutagenog potencijala monoterpena kamfora, eukaliptola i tujona i ispitivanje molekularnih mehanizama njihovog bioantimutagenog delovanja predmet njene doktorske disertacije. Nakon odbrane doktorske disertacije i izbora u zvanje docenta, nau?no-istra?iva?ki rad dr Biljane Nikoli? se pro?iruje na ispitivanja razli?itih biolo?kih aktivnosti prirodnih proizvoda u prokariotskim i eukariotskim model sistemima (antimikrobna, citotoksi?na i genotoksi?na/antigeno-toksi?na), kao i biokompatibilnosti sinteti?kih materijala. U saradnji sa Fakultetom za fizi?ku hemiju Univerziteta u Beogradu dr Biljane Nikoli? se bavila i istra?ivanjima uticaja mikrotalasa na reprodukciju bakteriofaga u ?eliji doma?inu. Pored toga, u saradnji sa kolegama sa Instituta za zemlji?te u Beogradu, koleginica Nikoli? u?estvuje u istra?ivanju zemlji?nih bakterija koje podsti?u rast biljaka (plant growth promoting rhizobacteria). Rezultate svog dosada?njeg nau?no-istra?iva?kog rada dr Biljana Nikoli? je objavila u 23 nau?na rada, od ?ega 16 iz kategorija M21, M22 i M23. Imala je 65 saop?tenja na me?unarodnim nau?nim skupovima, od toga 8 ?tampanih u celini, kao i 12 na nacionalnim nau?nim skupovima, od toga 1 ?tampan u celini. Navedene bibliografske kategorije, zajedno sa magistarskom tezom i doktorskom disertacijom, ?ine zbir od 102 bibliografske jedinice. dr Biljana Nikoli? je do sada bila u?esnik na 4 nacionalna i 1 bilateralnom nau?nom projeku. (2015) Characterization of probiotic properties of human strains of Lactobacillus plantarum, 10th Serbian Microbiology Congress-MIKROMED, Beograd, Book of Abstracts ISBN 978-86-914897-2-4, 223.ANALIZA NAU?NOG RADAPrema problematici koja je izu?avana, radovi dr Biljane Nikoli? se mogu podeliti u nekoliko celina. Najve?i broj radova bavi se problematikom antimutageneze/antikancerogeneze, dok je u manjem broju radova ispitivan ?iri spektar biolo?kih aktivnosti prirodnih proizvoda, biokompatibilnost stomatolo?kih materijala, kao i efekat mikrotalasa na reprodukciju bakteriofaga u ?eliji doma?inu. Po jedan publikovani rad bavi se ekogenotoksikologijom, probiotskim svojstvima izolata roda Lactobacillus, kao i mikrobiologijom zemlji?ta.Radovi iz oblasti antimutageneze/antikancerogeneze su ozna?eni brojevima 1.2., 2.1., 2.2., 2.3., 2.6., 3.1., 3.2., 3.4., 4.2., 4.3., 5.1. i 5.2. Pored toga, rezultati dobijeni u ovoj oblasti ?tampani su u celini i u saop?tenjima 6.1., 6.3., 6.4., 6.5., 6.6., 6.7., 7.1. i 8.1., a tako?e i u brojnim saop?tenjima sa me?unarodnih i nacionalnih skupova ?tampanih u izvodu. Svi pomenuti radovi mogu se podeliti u dve velike grupe: one koji ispituju antimutagenost antioksidanata i one koji ispituju bioantimutagene. Istra?ivanja antimutagenog efekta etarskog ulja bosiljka (Ocimum basilicum L.) i njegovog dominantnog konstituenta linalola prikazan je u radu 2.1. Dobijeni rezultati primenjenih reverznih testova na Escherichia coli K12 i WP2 sojevima, kao i komet testa na kvascu Saccharomyces cerevisiae, ukazuju da pomenuti antimutageni inhibiraju oksidativnu mutagenezu, te da se ovo njihovo svojstvo mo?e povezati sa antioksidativno??u. U radu 2.2. prikazani su rezultati istra?ivanja antimutagenog potencijala terpenoida linalola, mircena i eukaliptola prema oksidativnoj mutagenezi; ispitivanje je vr?eno primenom reverznog bakterijskog testa na E. coli WP2 oxyR, kao i primenom komet testa na humanim hepatoma (HepG2) i limfoblastoma (NC-NC) ?elijama. Efekat metaboli?ke transformacije S9 frakcijom na antimutageni potencijal pomenutih monoterpena prema t-butil hidroperoksidu i 2-nitropropanu ispitan je reverznim testovima na E. coli WP2 i Salmonella typhimurium, ?to je prikazano u radu 4.3. Isti monoterpenoidi bili su predmet istra?ivanja i u radu 5.1, u kome je ispitan njihov potencijal da u komet testu na humanim hepatoma ?elijama inhibiraju genotoksi?ni efekat kancerogena iz hrane IQ i PhIP. Antimutagenim potencijalom model antioksidanata vitamina E i C bave se radovi 3.1. i 3.2. U radu 3.1. u bakterijskim reverznim testovima ispitan je antimutageni potencijal vitamina E prema oksidativnoj mutagenezi. Efekat vitamina C prema oksidativnoj mutagenezi, prikazan u radu 3.2, obuhvata rezultate dobijene u reverznim testovima na E. coli i S. cerevisiae, kao i u komet testu na S. cerevisiae. Grupi radova koji se bave antigenotoksi?nim potencijalom antioksidanata treba dodati i saop?tenje ?tampano u celini 6.1, koje se bavi antimutagenim efektom frakcija ?alfije (Salvia officinalis L.) detektovanom u bakterijskim reverznim testovima. Antigenotoksi?ni potencijal ekstrakata biljaka iz roda Allium (A. melananterum i A. flavum), koji se tako?e mo?e pripisati njihovim antioksidativnim svojstvima, prikazan je u radovima 2.6. i 6.5. Antigenotoksi?nost ekstrakata Allium sp. ispitana je reverznim testom na E. coli WP2 oxyR soju, koji je posebno osetljiv prema oksidativnoj mutagenezi, kao i alkalnim komet testom na humanim fibroblastima plu?a. U radu 2.6. pored antigenotoksi?nosti ekstrakata, ispitan je i njihov efekat na sintezu i aktivnost antioksidativnih enzima katalaze i superoksid dismutaze u humanim fibroblastima plu?a. Drugu veliku grupu radova iz oblasti antimutageneze ?ine oni koji se bave antimutagenim potencijalom bioantimutagena iz etarskih ulja ?alfije i bosiljka. Bioantimutageni potencijal etarskog ulja ?alfije (Salvia officinalis L.) i njegovih frakcija, uo?en je u eksperimentima UV-indukovane mutageneze pomo?u reverznih testova na E. coli K12 i S. cerevisiae D7, ?to je prikazano u radu 7.1. Radovi ozna?eni brojevima 2.3. i 3.4., kao i saop?tenja ?tampana u celini 6.3., 6.4., 6.6. i 6.7, bave se antimutagenim potencijalom monoterpena iz etarskog ulja ?alfije - kamforom, eukaliptolom i tujonom, ?to je bio predmet doktorske disertacije kandidata. U radovima je prikazana dualna priroda testiranih monoterpena, ?to je bilo osnova za obja?njenje molekularnog mehanizma uo?enog antimutagenog dejstva prema UV zra?enju i UV-mimetic agensu 4NQO. Naime, iako genotoksi?ni, monoterpeni primenjeni u malim koncentracijama indukuju malu koli?inu DNK o?te?enja i zahvaljuju?i tome stimuli?u mehanizme reparacije DNK koji ne gre?e, uklju?uju?i reparaciju isecanjem nukleotida, rekombinacionu reparaciju i reparaciju pogre?no sparenih baza. ?elija u kojoj je nivo odbrane od DNK o?te?enja na ovaj na?in podignut, ima pove?an kapacitet da ispravi i o?te?enja izazvana model mutagenima UV i 4NQO, te zahvaljuju?i tome monoterpeni u malim koncentracijama deluju antigenotoksi?no i antimutageno prema indukovanoj mutagenezi. Ovakvo protektivno delovanje malih doza monoterpena u saglasnosti je sa u toksikologiji prihva?enom teorijom hormezisa, kao ?to je obja?njeno u radovima 2.3. i 3.4., kao i u saop?tenjima 6.6. i 6.7.Pored toga ?to etarsko ulje bosiljka (Ocimim basilicum L.) ima potencijal da redukuje oksidativnu mutagenezu, ?to je prikazano u radu 2.1., ono ima sposobnost da smanji i u?estalost UV-indukovanih mutacija, kao ?to se vidi iz rada 5.1. Koriste?i reverzne testove na E. coli K12 i S. cerevisiae D7, pokazano je da etarsko ulje bosiljka i njegov dominantni sastojak linalol smanjuju u?estalost UV-indukovanih mutacija i to zahvaljuju?i pove?anju efikasnosti reparacije isecanjem nukleotida. Radovi 1.2., 4.2. i 8.1 prikazuju antimutageni potencijal obe grupe prirodnih jedinjenja – antioksidanata i bioantimutagena. U radu 4.2. ispitivan je antimutageni potencijal monoterpena kamfora, eukaliptola, tujona, linalola i mircena prema mutagenezi indukovanoj UV zra?enjem i model mutagenima 4NQO i t-BOOH, a efekat je pra?en u bakterijskim reverznim testovima na E. coli K12 i WP2 sojevima. Monografija 1.2. i saop?tenje sa nacionalnog skupa ?tampano u celini 8.1. prikazuju ciljeve studija antimutageneze i obja?njavaju molekularne mehanizme delovanja terpenoida iz ?alfije i bosiljka. U grupu radova iz oblasti genotoksikologije mo?e se priklju?iti i rad 1.1. iz oblasti ekogenotoksikologije, koji prikazuje efekte genotoksi?nih polutanata u ?ivotnoj sredini, isti?e zna?aj ekogenotoksikolo?kog monitoringa, ali i supstanci prirodnog porekla koje poseduju antigenotoksi?ni potencijal.Deo istra?ivanja kandidata odnosi se na ispitivanja ?ireg spektra biolo?kih aktivnosti prirodnih proizvoda. U radu 3.3. hemijski je okarakterisano etarsko ulje Thymus capitatus, a zatim su prikazana antioksidativna, citotoksi?na i antimikrobna svojstva ulja i dominantnih sastojaka karvakrola i timola. Citotoksi?nost etarskog ulja je pra?ena primenom MTT testa na kancerskim ?elijama kolona (HCT 116 i HT-29), kao i na normalnim fibroblastima plu?a (MRC-5). Antimikrobni efekat je ispitan primenom ve?eg broja bakterijskih i fungalnih sojeva iz ATCC kolekcije. U radu je prikazan i efekat ulja i dominantnih konstituenata na adheziju E. coli i L. monocytogenes na ?elije kolona HT-29. Radovi 5.3. i 5.4. bave se ispitivanjem antimikrobnog potencijala prirodnih proizvoda; u prvom su prikazani rezultati optimizacije mikrodilucione metode za ispitivanje antibakterijskog potencijala, a u drugom antibakterijski efekti etarskih ulja Juniperus sibirica i Juniperus sabina. Protektivni efekat vinskih marinada sa ekstraktima za?inskih biljaka na razvoj mikroorganizama-patogena hrane prikazan je u saop?tenju ?tampanom u celini 6.8. Uo?en je inhibitorni efekat me?avine crnog vina Merlot i ekstrakata ?alfije, nane i kima na rast L. monocytogenes i S. aureus u uzorcima svinjskog mesa. U radovima 2.4. i 2.5. prikazana su istra?ivanja biokompatibilnosti novih endodontskih nanostrukturnih materijala, proizvedenih primenom hidrotermalnog sol-gel procesa, a koji su po hemijskoj strukturi aktivni sistemi silikata i hidroksiapatita. Ispitivana su njihova fizi?ko-hemijska svojstva, citotoksi?nost prema humanim fibroblastima plu?a (MRC-5 ?elije), genotoksi?nost u humanim limfocitima periferne krvi, kao i imflamatorni efekat u histopatolo?kim studijama na mu?jacima Wistar pacova. Dobijeni rezultati preporu?ili su nove endodontske nanomaterijale za budu?a klini?ka ispitivanja, a u cilju njihove potencijalne primene u stomatolo?koj praksi.Radovi 4.1. i 4.4. i saop?tenje ?tampano u celini 6.2. prikazuju rezultate istra?ivanja biolo?kih efekata mikrotalasnog zra?enja na modelu umno?avanja bakteriofaga λ u doma?inu E. coli K12. U pomenutim radovima autori uo?avaju da efekat mikrotalasnog zra?enja frekvencije 2,45 GHz na umno?avanje λ faga zavisi od temperature - u eksperimentima sprovedenim na temperaturi bliskoj optimalnoj (37°C) mikrotalasi usporavaju proliferaciju faga, ali na sub-optimalnoj temperaturi oni izazivaju suprotan efekat i ubrzavaju razmno?avanje faga. U analizi mogu?ih mehanizama delovanja mikrotalasnog zra?enja razmatraju se i njihovi termalni i netermalni efekti, a pretpostavlja se da uo?eni efekat uklju?uje pove?anu ekspresiju heat shock proteina.U radu 4.5. prikazan je rezultat karakterizacije novih izolata bakterija roda Lactobacillus. Izolati imaju humano, oralno ili fekalno poreklo. Fenotipska i genotipska identifikacija su pokazale da se radi o sojevima L. plantarum, L. casei i L. paracasei. Ispitivane su njihove hemoliti?ke i antimikrobne aktivnosti, sposobnost agregacije, potencijal adhezije na hidrofobni rastvara? heksadekan i na ?elije kolona u kulturi (HCT 116), tolerancija prema lizozimu, niskim pH vrednostima i ?u?nim solima. Pra?eno je i pre?ivljavanje sojeva u uslovima ve?ta?kog gastri?nog i intestinalnog fluida. Dobijeni rezultati ukazali su na dobra probiotska svojstva testiranih izolata.U radu 4.6. prikazani su rezultati ispitivanja potencijala pojedinih sojeva zemlji?nijh bakterija da pobolj?aju rast i resorpciju nutrijenata kod biljaka je?evica (Dactylis glomerata L.) i livadski vijuk (Festuca pratensis Huds.), ina?e zna?ajnih u proizvodnje sila?e. U eksperimentima in vitro i in planta je pokazano da sojevi rodova Azotobacter, Bacillus, Pseudomonas, Rhizobium i Shinorhizobium poseduju svojstva pobolj?anja rasta obe poljoprivredne kulture.Na osnovu analize radova mo?e se zaklju?iti da je nau?ni doprinos dr Biljane Nikoli? zna?ajan za oblast mikrobiologije, genetike prokariota i genotoksikologije, naro?ito mutageneze i antimutageneze.
