Applied Biosystems RapidFinder™ Express Software v1.1, SDS Software Version 1.4.2 and RapidFinder™ Express Pathogen Updater v1.0 Release Notes

Software Release Notes for:

• Applied Biosystems RapidFinder™ Express Software v1.1 (Applied Biosystems 7500 Fast Real-Time PCR System)

• RapidFinder™ Express Pathogen Updater v1.0 (Deploys Listeria Spp. Pathogen Add On)

Minimum computer requirements:

The minimum computer requirements for 7500 Fast System with RapidFinder Express Software v1.1 and SDS Software v1.4.2 are:

o Intel® Pentium III 1 GHz or equivalent processor

o 1 GB RAM

o 20 GB hard drive

o Microsoft® Windows® XP Professional operating system SP2

IMPORTANT! Do not run antivirus applications while RapidFinder™ Express Software v1.1 is running.

Computer resolution requirements:

Please make sure to set the Screen Resolution to a minimum of 1024 by 768 pixels with True Color (32 bit) from (Control Panel> Display Settings).

Installation of RapidFinder Express v1.1 and SDS Software version 1.4.2:

• Insert the RAPIDFINDER EXPRESS v1.1, 7500 FAST RQS CD into the computer.

• Follow instructions from the installation wizard to install RapidFinder™ Express Software v1.1 and SDS Software v1.4.2.

IMPORTANT! Check and replace the lamp after installation if lamp usage approaches 2000 hours. The installer does not give any warning if lamp usage approaches 2000 hours during the installation. The next run after the installation will be aborted if lamp usage has already exceeded 2000 hours.

• Perform the following calibrations:

a. 7500 Fast Real-Time PCR System (ROI) calibration

b. Background calibration

c. Optical calibration

d. Pure-dye calibration

• Perform an RNaseP verification run.

Upgrade RapidFinder™ Express v1.0 and SDS Software v1.4.1 Software:

• Insert the RAPIDFINDER EXPRESS v1.1, 7500 FAST RQS CD into the computer

• Follow the instructions on the screen to install RapidFinder™ Express Software v1.1 and SDS Software v1.4.2.

IMPORTANT! Upgrading from the previous version 1.0 using the installation wizard will preserve existing RFE v1.0 calibration file. Please ensure that all your calibrations are up to date including FAM, VIC, ROX and NED. If you did not already calibrate the Pure Dye NED while using previous version v1.0, proceed to perform NED calibrations via SDS Software v1.4.2 application.

Upgrade From an earlier version of SDS Software:

• Installing RapidFinder™ Express software requires uninstallation of the existing SDS software using “Add & Remove Programs” from Windows® Control Panel.

• After un-installation, insert the “RapidFinder™ Express Software v1.1 and SDS Software v1.4.2” installer CD and follow the installation wizard instructions.

Installation of Pathogen Updates using Pathogen Updater v1.0 CD:

• Insert the Pathogen Updater v1.0 Installer CD and follow the instructions from the installation wizard.

• This will add Listeria Spp. kit into existing RapidFinder Express v1.1 Software installation.

• Users can create run files using Listeria spp. as a target pathogen, the next time RapidFinder Express v1.1 software is run.

New Features in RapidFinder™ Express v1.1 Software:

• Version 1.1 supports an E. coli 0157:H7 assay with multiplexed.

• Lamp Usage Tracking

o If lamp usage exceeds or equal to 2000 hours, “Run Aborted” message will be displayed when a run is attempted. The run will not start if the lamp has been used for more than 2,000 hours.  Replace the lamp, either by contacting Applied Biosystems to schedule lamp maintenance, or by following the instructions in the Applied Biosystems 7300/7500/7500 Fast Real-Time PCR System Installation and Maintenance Guide.

IMPORTANT! - After the lamp is replaced all calibrations must be executed to ensure data accuracy.

If lamp usage is between 1750 and 2000 hours, the run will start but a warning message will be displayed at the end of the run as a reminder to plan for lamp replacement soon.

o If lamp usage is less than 1750 hours, all runs will start and complete successfully without a lamp usage warning or error messages.

• Print Support for A4 / Letter Size and Portrait / Landscape orientation.

• Results can now be printed in Run layout format under View Results->Report Tab.

• Improved performance response time while accessing and displaying existing runs.

Known Issues with RapidFinder™ Express v1.1 Software:

General Issues

• Do NOT save any RAPIDFINDER™ EXPRESS Run Files in the SDS Software. RapidFinder Express Run Files that are saved in SDS Software cannot be re-opened by RapidFinder Express software, and RapidFinder Express results will be invalidated.

• If you attempt to launch SDS Software version 1.4.2 while the RapidFinder Express Software is running, the SDS software will not start. SDS Software version 1.4.2 cannot be run at the same time as the RapidFinder Express software.

Entering Sample Names

• When you use the Fill Down feature, and your starting sample name has a number, RapidFinder Express software will automatically increment the number. The maximum number that can be used is 100,000. If a larger number is attempted, an error message will be presented. Also, Applied Biosystems does not recommend using copy and paste operations in conjunction with this “auto-increment” feature.

Creating a Run File

• If you click Close while on the “Enter Run File Information Page” page, the settings on this page will not be saved. Settings will be saved only after user proceeds to the “Confirm Run Layout” page.

Creating a New Template from a Run File

• The maximum width of the “Creator” field is 22 characters. Text longer than 22 characters will be truncated with ellipses (…).

Pipetting Samples

• If fewer than 8 tubes are needed, the 8-tube strips can be cut. However, the Optical 8-Strip cap cannot be cut. The entire Optical 8-strip must be used to close the tubes.

Instrument Run

• During the run, on the “Start Instrument Run” page, the information in the “Cycle Counter (Cycle n of n)” and “Time remaining” fields may not be updated in real-time. However, the run will complete successfully.

• When an instrument run is in progress, the RapidFinder™ Express v1.1 Software requires significant CPU utilization to perform data collection activities. In order to allow the workstation the greatest processor usage, Applied Biosystems recommends avoiding the following activities during an instrument run:

o Accessing the Internet;

o Writing to a CD or DVD;

o Scanning a disk for viruses;

o Defragmenting a disk

o Any other activity that requires extensive CPU use

Performing such an activity during an instrument run can result in data loss or termination of the run.

Viewing Results

• For all “Negative” sample results, check for warnings under the “Results By Location” tab to ensure that there are no warnings associated with the sample. Samples with warnings may need further evaluation by a qualified analyst.

Known Issues in SDS Software v1.4.2:

Display Settings

The RapidFinder™ Express software requires display settings and graphics card capable of supporting 16-bit color palette. Computer displays with lower color palette settings with fewer colors will result in the incorrect display of some detectors.
