Procedure: Plasmid Engineering

Your assignment is to model the technique used to combine pieces of DNA. You will form a new plasmid that contains a piece of foreign DNA. The new plasmid is called recombinant DNA. A new segment of DNA can be attached to a plasmid only if its exposed bases (sticky ends) are complimentary to the exposed base on the DNA of the plasmid. The plasmids represented on the page titled Plasmid Engineering have been chemically cut by restriction enzymes at the nucleotides indicated by dashed lines. Your assignment is to complete the genetic engineering process.

1. Cut out the figures on the bottom of the page titled Plasmid Engineering.

2. Label the blank boxes in Segments A and B to show the base sequence necessary to attach them to the complementary plasmid.

3. Use glue or tape to attach the DNA segments to the corresponding plasmids. Show your teacher.
